I. Product Introduction
Fluorescently labeled Annexin V is widely used to detect the externalization of phosphatidylserine (PS) on the plasma membrane of cells undergoing early apoptosis. However, adjusting flow cytometry compensation can be challenging in certain experimental settings. This reagent facilitates the binding of fluorescently labeled Annexin V to PS, thereby simplifying and streamlining the compensation adjustment process.
II. Operating Steps
1. Collect 5 × 105 - 1 × 106 The cells were centrifuged and washed twice with pre-cooled PBS, and the supernatant was discarded.
2. Resuspend in 500 μl of Apoptosis Positive Control Solution and incubate on ice for 30 minutes.
3. Centrifuge and wash with pre-cooled PBS, then discard the supernatant.
4. Set up two single-staining tubes. Following the apoptosis kit instructions, add an appropriate volume of pre-cooled 1× Binding Buffer to each tube to resuspend the cells, then stain with fluorescently labeled Annexin V or PI/7-AAD and incubate.
5. Adjust the voltage and compensation on the machine.
III. Save
Store at 4°C; stable for up to 12 months. Transport with ice packs.
