      Instructions for Use

      This procedure is suitable for processing samples ranging from 0.1 to 2 mL (up to 2×10⁸ cells), using FiniGet Fenbide® 140–2000 single-hole magnetic stands and 5 mL sterile flow cytometry tubes.

      Taking the sorted human PBMC NK cells as an example:



       

      Operating Procedure (Starting with PBMCs, Pouring Method) :

      1. Preparation of human PBMCs : Utilize Ficoll density gradient centrifugation PBMCs were isolated from human peripheral blood. The PBMCs were collected, washed with PBS, and after centrifugation, resuspended in sorting buffer at a cell density of… 1×10⁸ cells/mL 。

      · If cryopreserved PBMCs are used, the cells will form clumps after thawing. Prior to sorting, the sample should be incubated at room temperature in a 100 μg/mL DNase I solution for at least 15 minutes and then filtered through a cell strainer.

      · For the sorting buffer, we recommend using PBS containing 1 mM EDTA and 2% fetal bovine serum (FBS), which should be filtered through a 0.22 μm membrane filter to remove bacteria prior to use. The buffer must not contain calcium ions, magnesium ions, or biotin. Alternatively, you may also use FiniGet Fenbide® 140-2001 Cell Sorting Buffer.

      2. Pipette 100 μL of cell suspension (1×10⁷ cells) into the bottom of the flow cytometry tube, then add 10 μL Human NK Cell Isolation Cocktail , mix well, Incubate at room temperature for 10 min. 。

      · If sorting a larger number of cells, the amount of Human NK Cell Isolation Cocktail can be increased proportionally.

      3. Add 10 μL Streptavidin Beads (Shake vigorously for about 30 seconds before use; do not centrifuge at high speed after mixing.) Mix well, Incubate at room temperature for 2.5 min. 。

      · If sorting a larger number of cells, the amount of Streptavidin Beads can be increased proportionally.

      4. Add sorting buffer to bring the volume to 2.5 mL , after gentle mixing, place the flow cytometry tube into the magnetic stand. Let stand for 2.5 min 。

      · The experimental volume should not exceed 2.5 mL to prevent exceeding the magnetic separator’s sorting range.

      5. Pick up the magnetic holder, Pour gently and smoothly. The cell suspension is placed in a sterile centrifuge tube, with the magnetic poles and flow cytometry tubes kept in an inverted position. 2-3 s Subsequently, it returns to an upright orientation. This suspension contains purified human NK cells.

      · During the pouring process, the flow cell must remain attached to the magnetic stand.

      · Do not shake the tube or touch, via the tube opening, any droplets that may still be clinging to the flow‑cell inlet. Such droplets must be discarded.

      6. After washing the cells, resuspend them in the desired buffer or culture medium, and they are ready for downstream applications.
